Evaluating the Impact of Enzyme and Transporter Contributions to Oral Bioavailability: A Meta-Analysis
This meta-analysis provides a careful evaluation of toxicokinetic processes most likely to modulate oral bioavailability in humans, using human in vivo study data across 60 chemicals, and evaluating the contribution of hepatic clearance, CYP450 and UGT metabolism and transporter action. This analysis will enable the development of an approach to anticipate modulation of bioavailability in an efficient manner, and will be critical in informing next steps needed to modify existing higher throughput TK tools used in New Approach Method strategies for more efficient and predictive chemical evaluations.
